Put some oil back in it and see if it will run. After about 10 or 15 minutes, check for water in the oil. It is unlikely water will show in the oil that fast unless you have a cracked head or block. If you have access to a leak down tester, it will find the leak source. Remove all the spark plugs. Rotate the engine so the test piston is top dead center on the compression stroke. The leak down tester puts low pressure air into the spark plug hole. If you have a leak, you will hear the air escaping, like from the carburetor, adjacent spark plug hole, exhaust, crankcase, or the cooling system. I would start with the cylinder that had water in it. Good luck.
